Welcome back, everybody. Here's our next question. PC R is used to amplify a DNA sequence many times resulting in sufficient DNA copies for analysis. Which of the following techniques is used to visually evaluate the PC R reaction choice A Sanger sequencing choice B. Next generation sequencing choice C gel electrophoresis or choice D centrifugation. Well, an easy one to eliminate right away is choice decent. That's when you spin samples at very high speeds to separate components of a solution by density. And you don't use that to visually analyze PC R. So because all DNA will have the same density and you want to make sure and have confirmation that you have your specific DNA fragment that you've been amplifying and you would not do that by centrifugation. Then we can also eliminate choices A and B both of which are sequencing methods. This would give us the sequence of our PC R product um but it isn't giving us a visual way to evaluate that we have our amplified fragment. The difference is Sanger sequencing is the original technique. Next generation is a high throughput method where you have all sorts of DNA samples being run in parallel. This is what allowed us to finish the human genome project sort of the. Well, as it says, next generation most advanced sequencing method for large numbers of samples. But again, both of these are sequencing methods, not a way of visually evaluating the success of our PC R reaction. So we're left with gel electrophoresis choice C. And that is indeed our correct answer because the gel will show bands of DNA fragments, then you can see the band corresponding to our PC R product. And if you've amplified it successfully, you'll see a band of the correct size and you'll see it nice and dark that you've made a lot of that product, which is the goal of PC R.
