Magnification and Optical Path

Microscopes use a combination of lenses to magnify specimens. The total magnification is calculated by multiplying the magnification of the ocular lens by that of the objective lens.

  • Ocular Magnification: Usually 10x.

  • Objective Lens Magnifications: Commonly 4x, 10x, 40x, and 100x.

  • Total Magnification:

    • 4x objective × 10x ocular = 40x

    • 10x objective × 10x ocular = 100x

    • 40x objective × 10x ocular = 400x

    • 100x objective × 10x ocular = 1000x

  • Light Path: Light from the lamp passes through the condenser, then through the specimen, objective lens, and finally the ocular lens.

Proper Handling and Common Mistakes

Correct handling of the microscope is essential to prevent damage and ensure accurate results. Several common mistakes should be avoided:

  • Never place a slide under the stage clip: This can crack the slide. Incorrect slide placement under stage clip

  • Never clean the lens with paper towels: This will scratch the lens. Always use lens paper. Lens paper for cleaning microscope lenses

  • Stage Motion Knobs: Allow precise movement of the slide for scanning.

  • Correct Slide Placement: Place the slide gently on the stage and secure it with the clips, avoiding forceful placement.
